Jesus was very clear on this: The greatest commandment is to love the Lord with all that we are, and the second is to love our neighbors as ourselves. Seems to me, if those two commandments are obeyed fully, none of the rest could possibly be disobeyed.
It seems as though there are levels of judgment based on sins -- recall Jesus' statements to the cities that rejected Him (Matthew 11), which included that "it will be more tolerable for Sodom in the day of judgment than for you." If the day of judgment is more tolerable for one than another, then it makes sense that there is more severe or less severe judgment, hence -- I think -- greater and lesser sins.
Also, when Jesus was speaking with Pilate in John 19, He said that "the one who handed me over to you is guilty of a greater sin."
The word "sin" in Greek, as you may know, means "missing the mark." Implicit in the very etymology of the word is the idea that one can sin "more" or "less" -- i.e., be closer or farther from the mark.
I will never be convinced that a person who lies by telling his wife she's the most beautiful woman on earth has committed a sin of the same magnitude as a person who is proud of not being a sinner like others around him.

All the commandments were given as laws by God to prevent us humans from harming and destroying one another. A lot of them have to do, really, with the "Golden Rule" - don't lie, cheat, steal, have sex with someone else's significant other, don't want someone else's stuff to the point you will do evil to get it, and leave God out of your problems if He is not directly involved. If you have love in your heart for your fellow man, you will have no desire to do any of those things that will hurt another or yourself. There is no one commandment that trumps another - even murder (justifiable homicde comes to mind - I would forgive someone for murdering a child molester!). Through the Atonement of Jesus Christ, who took upon Himself your sins as well as everyone else's in the Garden of Gethsemane, we all have the chance to repent of all of the commandments we have broken, and sin no more.

Well, Jesus called the most important 2:
Love the Lord thy God.... and the second is like unto it, love your fellow men (and women!)..... On these 2 hang all the law and the prophets.
