Are you looking for free or commercial applications? Web based or desktop?

You can use applications like Google Earth pretty much for free for personal use. You can go out and purchase a high end application like ESRI's ArcMap or Intergraph's GeoMedia. If you are looking for web based mapping, a lot of open source mapping projects exist. Check out OpenMap or KaMap for free AJAX type mapping scripts. If you want commercial, ESRI has ArcIMS or ArcGIS Server and Intergraph has GeoMedia Server.

I am madly in love with mapquest.com.

The maps and instructions are printable, with alternate routes easily available.

i would go to yahoomaps.com and you can print up where you want to go. its cheap and free.
