First Class has more amenities, but Business Class is cheaper and gives the room. Cheaper includes when using airline miles.

As for redemption, it's going to vary from one airline to another--but for starters most will require you to have achieved a premeire status first, or will require you to "spend a bunch of points/miles" to get that upgrade.

First is better, but requires more. I find Business class to be good enough. There is a huge difference between Economy and Business, but not as much difference between Business and First.

You call the airline to do the upgrades. Every airline has different rules depending on the type of ticket you have purchased. You can request the upgrade at any time, but the actual upgrade may not happen until the day of your flight depending on availability. Again, best to look at the rules of the airline on its website or give them a call and ask about your specific flight.

You need to do that while buying or booking your seat. First class is of course better, since it offers more comforts and better selection of in-flight food and beverages as compared to Business class.
