Grammatically, "another mug of stiff drink" is jarring, although possibly not absolutely wrong. "Drink" is rather rarely used uncountably, and with "stiff" it sounds even less natural. You'd more likely say "a stiff drink".

Pragmatically, neither one is very likely, because the bartender would want to know what brand you're after. If you're not at a bar, at there's only the one bottle on the table, either sentence would be ok.

Health-wise you're in trouble too, if you're drinking the hard stuff by the mug.

I would ask for a Bailey's, myself. "Could you get me another Bailey's, please?"
My husband would ask for a single malt.
Both in a glass, mine with ice, his with water.

Well, 'stiff drink' is rather vague....as is liquor actually because there are many types of liqour.

Also, you would not say 'mug' because a mug is traditionally used for tea and you wouldn't have an alcoholic drink in a mug.

'Could you pour me another shot of ___?'
'Could you pour me another glass of___?'

Fill in the alcholic drink with whatever one you choose. Alternatively if you want beer or an alchopop like WKD then you can say:

'Could you get me another bottle of ? (or whatever beer/alchopop you drink!)

And don't forget the 'please'! =)

Could you pour me another mug, make it a double.

Liquor however, is not generally served in a mug.
