If the holiday was all about the trip getting TO Rome, then the former. If the time spent IN Rome was the essence, then the latter.

For the second sentence, they are nearly the same, and you could have also used the word "during."

I'd say "in" for both.
I might say "to Rome" if only part of the holiday took place in Rome and a lot of the holiday was getting there. I wouldn't say "to" if I flew to Rome.

i always find that deciding the correct prepositions or conjunctions or whatever these things are tends to be something which you have to decide instinctively and is on the very edge of that critical mass of knowledge known as fluency in a language
i think all four are correct as there are usually a couple of ways of saying something although with the last one i had to swallow hard to convince myself as it sounds counter intuitive

Both should be "in".
