Soviet cosmonaut Yuri Gagarin was the first man in space, making one orbit in Vostok 1 on 12th April 1961.
Vostok 2, carrying cosmonaut Gherman Titov was launched from Baikonur at 0600 UT on Sunday 6 August 1961.
Alan Shepard's spaceflight was suborbital. The first American to go into orbit was John Glenn, on February 20, 1962.
Vostok 3 and Vostok 4 were launched a day apart (August 11 and 12th 1962) , and together these missions marked the first time that more than one manned spacecraft was in orbit at the same time, giving Soviet mission controllers the opportunity of learning how to manage this scenario. Vostok 3 was manned by cosmonaut Andrian Nikolayev, Vosrok 4 had cosmonaut Pavel Popovich on board.
So it looks like John Glenn was the third man in space.

I really don't understand why so many people can't answer this question correctly. Some people even put the dates in and still name the wrong person as the third man in space.
Yuri Gagarin was the first (April 1961), followed by Alan Shepard in May 1961, followed by Gus Grissom in July 1961. Therefore, and this seems straightforward enough, Captain Virgil I. 'Gus' Grissom was the third man in space.
Of those three men Gagarin was the only one to fly an orbital mission, Shepard and Grissom making suborbital space flights. Gherman Titov flew in August 1961, and John Glenn flew in February 1962, both making orbital flights, so John Glenn was both the third American astronaut to fly in space and the thrid man ever to orbit the Earth.

Yuri Gagarin was the first person in space (12 April 1961), followed by Gherman Titov (6-7 August 1961). Alan Shepard was the first American on 5 May 1961 in the Mercury-Redstone flight Freedom 7.
As pointed out above, Shepard's flight was suborbital, however he reached an apogee (highest point) of over 116 miles. An altitude of around 100 miles is generally accepted as being the beginning of space, ergo Shepard was the third man in space.
Shepard was followed on 21 July 1961 by Gus Grissom in the Mercury-Redstone, Liberty Bell 7. John Glenn achieved orbital flight on 20 February 1962 in the Mercury-Atlas Friendship 7.
