Although sleeping is a good alternative, it might not be helpful when you're at work. First you need to know why your eyes are tired...if you work in front of a monitor then what you can do is stop after 30 minutes and close your eyes and roll your eyeballs in all directions possible, this way, you relieve your ocular muscles from the strain (this also helps if you are reading tons of things). If you feel that your eyes being tired is unusual then maybe you should see an Ophthalmologist (not an optometrist). You might need correction glasses. When your eyes are not in perfect condition meaning 20/20, your muscles are working triply hard to focus on the things you read. You cannot continually use your eyes without relaxing its muscles every so often.

Consider closing your eyes, covering them with a damp cloth for about 10 minutes.

If you have the time and opportunity, it's likely what you really need to do is get some sleep. You are fatigued.

If you don't want to fall asleep just rub the tops with your pointer finger. Don't dig at them though. When I wake up in the morning sometimes I have a bad habit of rubbing them too hard. Then they hurt really bad all day! Bona Fortuna(GOOD LUCK)!

Splash some cold water on them (not on the actual eye of course)...feels great, wakes you up, and will make them feel less irritated.
