If you're coming from Boston take the Masspike (I-90) west to I-84 west and follow I-84 through Connecticut and cut down at I-684 and take that all the way into the Cross County Parkway to the Thruway south and you'll end up in the Bronx and can access any of NYC from there.

Joe S hit it on the head. I live an hour north of NYC and drive to Boston a few times a year. If I go north and get on the Mass Pike around Albany, it's 5 hours or more. If I take 84 thru CT, it's 3 hours.
Depending on what side of the city you're staying on, do 90 to 84 to 684 and down.

90 to 84 to 91 to 95 is about 20 miles shorter, and typically 20-30 minutes faster than taking 95 straight there.

I have almost always used the 684 routing but you need to know that it does have two weak points in it
Hartford traffic and heavy precip
Try to avoid Manchester /Hartford / Waterbury a.m. traffic
By all means avoid the Hutch in any kind of heavy precipation; rain ,snow or sleet.
Instead go 287 -Port Chester -95 south
and then exactly where you need to go in Manhattan and which MLB team is playing at home will determine which bridge[s] you use. Directions to the upper west side will be very different than directions to the lower east side

the other answers using 90 to 684 to the hutch are good, but listen to the traffic reports every ten minutes, on AM radio 1010 wins on the ones, 1130 wbbr on the fives 88 wcbs on the eights

if you're gonig in the dead on night, I-95 can be faster.
if you're going between early morning and late evening, go the 84/684 route.
