My vote would go to Binions downtown because of the World Series of Poker finals being held there. Since that event has grown into what it is today, a good portion of the events and prelims are held at the Rio but Binions is still a classic.
Also famous is the MGM because of the events that are held there...especially boxing.
I guess famous is in the eye of the beholder.

bellagio is well known, but its more the hotel than the casino. lots of people flock to the hotel, and therefore also to the casino.
