Because back in the day, their primary jobs were to handle the cattle on ranches and drive the cow herd to market in far away cowtowns, such as Abilene, Kansas. And the cowboys would sing songs like:
"Whoopie--ti-i-o-o git along little dogies
It's your misfortune and none of my own.
Whoopie ti-i-o-o git along little dogies,
You know that Wyoming will be your new home."

Cowpokes are the guys at the stockyards who stand or sit along the top rail of the fence and prod (by poking) the cattle along as they are moved into or through the chutes to get branded, board the cattle cars of a train, whatever.
