Not really, I would say "There are four people in Tom's family. They are Tom, his father, his mother and his sister."
Also acceptable but a bit odd would be "There are four people in Tom's family: the father, the mother, the sister, and Tom." Normally we'd say "his" though, not "the".

"There are four people in Tom's family. They are his father, his mother, his sister, and Tom."
Or
"There are four people in Tom's family: His father, his mother, his sister, and himself."

Hmm I think you should say it by name, to say
"They are father, mother, sister and Tom" is not correct or sound.
You can say "They include his father, mother, sister and himself" Or "They include Joe, Jane, Jill, and himself/Tom."
