Other than the literal "Don't tell me!" Or is that the only meaning?

As you say the literal translation is Don´t tell me.

It is very colloquial so it is difficult to give a translation to an expression like that

It would equal to "Is that so?" more or less.

According to context, it can mean something like "I don´t believe you" (not offensive) to "Wow" to "That´s hard to believe."

It can mean many things. It can be a surprised response like "You're kidding!", or "Really?, "Wow!...", etc...
