They probably generally do not know that they are related.
When they are little, they can obviously tell their litter mates from other dogs. But when the female is in heat, she will mate with any male she can get to. Dogs are programed to mate when in heat.
It is actually a term called "inbreeding" depending on their genetics, they can produce healthy puppies, or they could produce sickly puppies.

Generally they know that they are related, but sometimes it won't stop them from breeding one another.. It's natural, it's hormone related, instinct.. That's why your pets should be spayed/neutered.. Then you don't have to worry about stuff like this.
