yes it does .. because in a shot of espresso there are fewer coffee beans used than in a cup of regular coffee .. and in fact.. the milder the coffe the more caffine you will get.

Yes. Espresso is brewed with boiling water (212F) which hinders the release of caffeine. Regular coffee is brewed with about 190F water (this temperature is called 'smile') and is the optimal temperature for caffeine release.
