Nice question! If your 2 year old brother is like mine, ignore him, until he finds out it doesnt work to annoy you. Thats what i do all the time. But, if he hits hard... spank him lightly on the hand and then let him cry for a while. After a while, he might want a hug, so give him a hug. then all of it stops for a while!
PS: This is advice from a 12 year old which has a 9 year old brother, and a 2 year old brother

A smack on the fanny. Then sit him in a corner for 15 minutes. Explain what you are doing and why. Let him know next time he will be in the corner longer. He is old enough to be taught right from wrong and should already know this. The talking is the most important part of this. Tell him it hurts his brother and makes him sad when he gets hit. Follow through.

Unfortunately, siblings fight. There is no stopping it. I have found out that if you let them fight it out it will lighten up.
It really depends on the ages of the children here. Obviously a baby isn't going to hit back his two year old brother. Then you would need to explain that we don't hurt people.
Eventually they will learn that they can't hit someone without any consequences. Hopefully they will get tired of being hit back and realize that they are getting hit b/c they hit.
Good luck!!
I know this method works wonders with biting.

Time-out, you can't stop them from hitting if you use spanking, don't get me wrong, if it warrents a spanking do it, but for last resorts, time out and corners do it for mine I have 3 boys and they and if they hurt each other they have to make who ever got hurt feel better, thats hard with a two year old but I make my 20 month old do it if he hits the 5 or 8 yr old, or a stern "no hitting" and strait to time out but not for 15 minutes maybe 5 with they forget what they've done in about 5 minutes
