Dr. Edward Jenner. The first helpful vaccine was created in 1796 by Edward Jenner. He injected an eight year old boy with the liquid, pus, or scabs from a patient of cow pox. The boy suffered from mild symptoms. Seven weeks later, Jenner vaccinated him with smallpox liquid. The boy didn't get smallpox so the vaccine had worked.The word vaccination derived from such creation. The term vaccine comes from a medical name for cowpox, vaccinia, which comes from the Latin word vacca, which means cow.

Some suggest the process that Jenner used to "create" the first smallpox vaccine was an ancient practice in many Asian cultures and was actually something he had witnessed as a child. He did make the milk-maid conection and put "two and two" together...but it is arguable that he swiped the idea of innoculation from somewhere in ancient Asia. A brilliant man though with multiple contributions!

According to Wikipedia, it is Dr. Jenner indeed. Pasteur invented the first vaccine against rabies, but Jenner invented the first vaccine ever - against smallpox

Well, Jenner didn't really create anything. He observed that milkmaids who had contracted cowpox (a mild related virus) were not getting sick in the smallpox epidemic. He then innoculated a small boy with cowpox and later exposed him to smallpox and he did not get sick.
