Seriously... Just tell them how you feel. That is the only way to go. It may not work out, but if you want anything more you need to just man up and communicate.

Its possible they do already. I dont know about other guys but when I have female friends, I have to make them appear in my mind unobtainable and not someone I would go out with in order to feel confortable around them. This conditioning helps seperate friends from potential girlfriends.
Of course in this situation it may be difficult. But since you are already his best friend it means its much easier to approach him and just be honest (depending on how you work your friendship). He will probally understand and work out if he wants to proceed or keep the friendship.
