Peanut butter, chunky, smeared on some old bread or a flour tortilla.
Be sure to chum the area to get Mr. Squirrel and his pals to be coming back again and again. I'd start chumming a couple of days before you put the trap out, spreading little pieces around the area. then when you put the trap out chum again. Put a big juicy piece on the trigger and hopefull you'll have one.
Fill up your garbage can with water and just dunk the whole trap in until he's drowned. Then scoop him out and toss him.
If you are thinking of letting him go in the woods somewhere....know that the little tree rats are like homing pidgeons....they will show back up in a couple of days.

I would put a mannequin dressed as a deer hunter in your live trap. No red squirrell alive can resist the temptation to go and chatter at a deer hunter. I bet you'll catch at least two dozen that way.

You all ready have the 3 best answers
DJ, Broker M, Dave G.
I start off with Peanut butter, chunky, and if they get use to that I switch to whole kernel corn.
Bird seed also works well.
This is the order I use them.

Bird seed hands down. I've caught more than my own share using bird seed.

Buy some really expensive flower bulbs. They seem to like those a lot. :)
