I'd say Astoria Queens rather than Brooklyn. If it's just got to be in Brooklyn, anywhere along the L train line would be pretty easy, obviously the closer to Manhattan the better. It's a straight shot crosstown on the L where you can get off at Union Square, 6th Ave or 8th Ave to transfer to get anywhere in Midtown. The L train is getting increasingly crowded though, especially at rush hours because of the gentrification explosion over the years. More condos are being put up every day. You didn't mention what you could afford in rent so I didn't take that into account.

I would say Brooklyn Heights which overlooks the East River and New York Harbor. The neighborhood is very quaint and has very nice houses and tree-lined streets. The commute is only about 20-30 minutes by train or if you are physical fitness minded - you could walk/jog/bike over the Brooklyn Bridge.

Park Slope or Brooklyn Heights. Both very safe, with lots to do. Park Slope just a little more affordable though.
