I've lived in FL primarily since 1972 and it is an expensive state to build/live in, certainly in/near those cities/areas most popular with tourists.
I live in South Central FL at the moment and in a developing, rural location. Large homes close to me are selling for $350 K UP. Actual cost to build may range from $175 per sq. ft. to $300 per sq. ft... The range is largely dependant on location, and availability of materials.
As one answer states, procuring the land is the logical first step, and that too will depend on location, with regard to it's price. Rural land is currently selling for approx. $25 per acre and UP. A zero lot line parcel within the city limits of Orlando, that will accomodate a 2000 sq.ft. house might sell for $75 K? UP. In 92 I had a 2000 sq. ft house, built on a zero lot line within the city limits, at the exterior. The lot in 92 as part of the cost, was $43 K. The total cost was close to $150. Today that house is market valued at near $400 K.
You should know the municipality and code regulations. You should know that where you build should be within a reasonable commute or delivery point to suppliers. With all due respect "Build Yourself" should allow for multiple skills, IE: Concrete work for a slab etc./Frame or block knowledge or experience/Utilities installation issues/Roofing, insulation, and various finishing issues.
You should be aware that no matter who builds, permits have to be pulled, and procedural inspections performed.
My usual advice to a DIY (while applauding them for wanting to do) is to let licensed/bonded/insured trades people do the major work. That doesn't say you have to contract with a major builder; you can sub out much of the work yourself. My rationale is that in issues of liability and insurability, the Pros, will be responsible, and I mention warantees. Once a shell is up, plumbing and eletcric in, roof complete, insulation installed, perhaps windows, THEN the DIY can finish the interior and exterior to suit themselves.
The major reason I suggest this is that anything a DIY does, that is assumed to save money in a project like this, might come back and BITE them at a later time.

hun its really expencive in Fl since i used to live there i can tell you that you would have to buy land before you can build anything the cost range of land is about 23 thousand to a couple million dollars for land this is what i have seen while living there for most of my life. then building the house could be from around 10 thousand or more depending on what type of house you want to build and how you want it look like etc. it also could be less. that is just a range.

Florida is expensive. Very expensive. This past year insurance rates have gone up about 80% and taxes aren't far behind. Land has skyrocked and so on. We've had a lot of friends this past year pack up and move out of state because they just can't afford it anymore. People that have lived here for years. And to build impact fees have gone sky high also. So think it over very carefully before moving here. Before you decide to build, maybe come down and rent for a 7 month lease. (6 month leases aren't available in Florida)

think hard before doing this. if you have money-then great! our homeowners insurance and property taxes are going thru the roof right now. land and building materials are very expensive due to our 2004 hurricanes and exporting concrete to China-i kid you not on that. you will need to find a realtor for the land and then a contractor. go online and start getting some estimates. good luck!

all of the answers on here are very accurate ive lived here for 15 years and i tell you its going up,up,up hardly affordabe anymore and to build is astronomical insurance, taxes. land etc are ridiculous we bought are house 8 yrs ago for 72,000 and now its apraised for over 300,000 are mortgage property taxes and insurance went sky high after the hurricanes so if your making really really great money and can afford it i recommend it but all of us here probrably wish we werent about now......
