Details the Chinese Civil War. But for a quick answer:

Chiang Kaishek had ultimate power in his grasp in the 1920's and 1930's, though he was challenged by Japan during their invasion in China's War of Resistance.

It is widely though that he lost the war due to a series of economic and military blunders. Mao Zedong and his Communists took up geurilla warfare against Chiang and his nationalists and eventually overpowered them.

The Nationalists were forced to the island nation of Taiwan, while Mao and the Chinese Communist Party (CCP) decalred the People's Republic of China (PRC) in October of 1949. This ultimately turned out to be a blessing in digsuise for Chiang, as he turned Taiwan into an economic powerhouse.
