It should be said "He doesn't mean any harm" but these days it almost doesn't matter. Nobody really speaks using formal English.

The correct way is "he doesn't". The movie was probably showing someone using slang, or trying to show they were uneducated. Or it could be subtitle error, that happens. English is a living language and what's considered right changes over time, but for now "doesn't" is the right way.

You can, but it makes you sound kind of stupid.
Don't means Do not.
The correct way to say it would be DOES not, or Doesn't.
There's a second thing wrong with that sentence too.
It SHOULD say "He doesn't mean ANY harm."
Because if you say "He doesn't mean no harm." it means he does mean harm.
Like a math equation:
- -10 = 10.
It's called a "double negative".

As others have stated, it should be "he doesn't mean any harm". "He don't mean no harm" is slang, often used by uneducated or those of certain regions in the US. It's not an uncommon phrase in some southern states.

The proper way to say it would have been " he doesn't mean any harm"
but everybody uses slang while they talk. mind you, if you have to write that sentences down in proper English, it would be "He does not mean any harm"

Correct, but in accent it may differ. Doesn't is correct in this way, but also in songs it happens that they use he don't ... or she don't .... but it is more a way to slightly differ the feeling of the scentence than anything else.
