the system may be sized correctly, however, it sounds like the design is bad. There should be volume dampers from the sounds of it so that flow is restricted where close so that it flows further to other rooms.

To truly have it checked, it would need to be reviewed by a mechanical engineer which will be pricey.

All the ducts should have adjustable dampers in them somewhere near where they are taken off the main duct.

First make sure that all of the dampers are fully open, then try to partially close the dampers in the ducts to the rooms that have sufficient cooling until you are able to get air flow into the room under question.

This is just simply balancing the system. Partially closing the dampers on the ducts for which there is sufficient air flow will not affect the flow of cold air to those rooms by very much, and can make a big difference in air flow into the warm room.

You could check your air intake filters are clean. If you closed all the other rooms off and you still got no flow then your furthest room must have a restriction somewhere.
Remove ceiling panels and test drill through the ducts till you find where the restriction is.

Btu output of 30 btu / sq.ft. should be enough. A 6" diameter duct can handle 150 sq.ft.

Check if the duct got crushed at some point.
