Man is separated from God because of sin. Fellowship was broken and the wages of sin is death. Man could not buy his way to righteousness through good works, deeds or rituals. The payment was death. But God provided a loophole, that a substitution could be made. This is where the sacrificial lamb became necessary. Your sins were passed onto the lamb, and the lamb took your payment for sin.
This was a foreshadow for He who was to come. For as Abraham said, "God will provide Himself a Lamb." This was seen when John the Baptist pointed to Jesus and proclaimed "Behold the Lamb of God that takes away the sin of the world."
If we ask Jesus to be Lord and Savior, we are, in essence, passing our sins onto Him as our personal lamb. Our payment was paid in full on the cross. Which is why Jesus said "Tetelestai" or "paid in full" (commonly translated as "it is finished").
So, with sin paid in full, we now have access to the throne wherein we can cry Abba, Father to the Almighty God. It is through Christ that our sins have been removed and thrown into the deepest ocean and as far as the East is from the West.

If I had the pick out the most profound thing that the Death of Jesus Christ illustrated, I would say that God Himself loved us so much that he Himself came after us.
We are a race of people who were dead to God. Something that He will live without forever, unless He decided to turn it all around and provide physical and spiritual healing to those who had no independent will to be his people.
He brings a spiritual life where there was only death. He countered sin with a covering atonement. He took whatever the keys are of Death, Hell, and the Grave, things that Satan held on to before He came. Jesus has the power to alter human history to His thought on what is good.
So the spiritual significance in this light ( I really don't believe that there is only one answer to this), is the authority He took from both man and devils, in order to regain the eternal life that man had lost long ago.

Jesus is the way! Jesus took our sin upon Himself (2 Corinthians 5:21). Jesus died in our place (Romans 5:8), taking the punishment that we deserve. Three days later, Jesus rose from the dead, proving His victory over sin and death (Romans 6:4-5). Why did He do it? Jesus answered that question Himself, "Greater love has no one than this, that he lay down his life for his friends" (John 15:13). Jesus died so that we could live. If we place our faith in Jesus, trusting His death as the payment for our sins - all of our sins are forgiven and washed away. We will then have our spiritual hunger satisfied. The lights will be turned on. We will have access to a fulfilling life. We will know our true best friend and good shepherd. We will know that we will have life after we die - a resurrected life in heaven for eternity with Jesus!

I thought I couldn't add to either makemeas… or Schneb's answers, but then I thought of this...
At the fall of man (Adam and Eve's disobedience) they immediately were aware of their sin and wanted to hide from God. Because of this, they wanted to be covered. As a covering for their bodies, and also a metaphorical covering for their sin, God himself killed an animal (that He created) in order to cover them with it's skin. It must have been grievous for God to do that.
God knew that there was no way that man could reconcile himself back to God (knowing what sin was and all) so God set in motion His plan for reconciliation. It had to be himself - in the body of Jesus - as a perfect sacrifice to redeem mankind.
Jesus accomplished that on the cross. He said "it is finished". That means that the ulitmate sacrifice had been made for us. He rose again, thus triumphing over death, and those who believe get to spend eternity with Him in heaven.

This is the most powerful event in human history.
God Himself, out of love for humanity came down and took human form and took on all our sins and the things that separate us from Him and bore them on His own(temporary) human body. He accepted the death of a criminal for our sakes and took our sins with Him to the grave.
Further, proving that He conquered all this He rose from the dead and promised that we also may be resurrected from the dead.
