I would want the dry rub on their for at least two hours, so if you're smoking for longer than that, no reason to put it on early unless you feel like it.

Depends on how much flavor you want. if you let it sit, then the rub will be sucked in by the meat. if you don't want the taste of the rub all the way through the meat then put it on right before smoking it.

Apply the rub right before putting it in the smoker if the rub has salt. The salt will draw out the moisture from the meat as it sits.
