It depends on the context.

upon = sobre, encima
he leaped upon his horse = saltó sobre su caballo

also en el momento de, inmediatamente después
upon receipt = al recibo, a vuelta de correo
upon the first opportunity = a la primera oportunidad

also contra
to turn upon him = volverse contra él

and also many different spanish prepositions like:
con : upon this terms = con estas condiciones
depend upon it = cuente con ello
de : to live upon grass = alimentarse de yerba
to depend upon one = depender de uno
etc.

"Upon" can mean various different things in English, and you didn't give an example of which meaning you wanted. But in most cases, "upon" means "on top of", so "sobre" is correct.

Here are some other ways to use "upon" that mean different things:
It came UPON a midnight clear
Once UPON a time
I wish UPON a star

None of those would use "sobre"

However it depends on the context.
"upon request" is "a pedido"

Its "sobre". If in English you were to say "It was put upon me" in Spanish it would be "Fue puesto SOBRE mi". Hope this helps.
