why is it that multiple-gene defects don't have a high chance of being cured by gene therapy?

The goal of gene therapy is to replace the gene that causes a disease or a defect with the normal gene. The problem with multiple gene defects is that both genes would have to be replaced and mostly likely it would have to be done at the same time.
