The problem here is going to be your layering of the graphic and the video, as they will essentially need to be on top of each other.
If you have PPT 2003, you could have the video on the slide you're working from, but perhaps behind the other graphic, and you'd have to add an exit animation for the graphic (perhaps just disappear, as this is quickest).
If you have an earlier version, you can run the video, but you're likely to have problems with the graphic being on top of the video (or else you'd have to have the video on top, in which case, you wouldn't be able to see the graphic in order to click on it at the beginning!). Having the video on the next slide (not a new window) may be the best option in this case.
So, here's how you do it:
Select the slide you want to work with. Go to the "Insert" menu, and choose "Movies and Sounds", then "Movie from File". PPT should ask you if you want the video to play "automatically", or "on click". If you're going to have the video on a second slide, select "automatically", so that as soon as you move off slide one, it will start playing. If you're going to play it on the same slide where the graphic is located, select "on click" (you might actually have to click on the video, as opposed to the graphic, but thish should be easily accomplished). You will need to size the video, just as you would any other graphic, by clicking and dragging the corners until it is the size that you want to display.
Also, if you are creating this presentation on a computer other than the one it will be presented on, you will need to send a copy of the video with the presentation, and re-establish the link on the presentation computer, or else PPT will be looking for it in the wrong folder.

Put an action button in your slide (slide show >> action buttons). A window with the header 'Action settings' will open. Click the radio-button in that window under "mouse click" tab against "hyperlink to". Then navigate to your saved video. On actual show, whenever you click the action button, the video will start playing automatically.

Powerpoint can only play MPEG and WMV video files in a slide. All other videos have to be played in another application.
