If you are the driver, might I suggest, new music on your Mp3 player, plenty of rest two nights before, plenty of nicotine if you smoke, favorite non-alcoholic beverage of your choice in conjunction with plenty of water and finally, the realization that the trip will take about 3 hours longer than anticipated. If you can leave in the middle of the night, DO SO!! I find that driving at night is MUCH easier since there is less traffic on the road. The drawback is that your body might not respond to being up at an unusual hour favorably.
If you are the passenger, music, beverages and a magazine will help pass some of the time, but be sure to look around and see where your're going and traveling through.

If you are driving, here is what I do. I try and get some sleep during the day and leave around midnight. That way when I start to get tired, then the sun is coming up and it keeps you awake. If I start to get sleepy, I leave a window partially rolled down, and take your shoes off and drive in your sock feet. If all this fails, try some hot coffee. It this fails then get off the road and get some sleep. don't push it, if you're having a hard time keeping your eyes open. I spent about 5 months in a hospital and about a year to recover after falling asleep at the wheel.

I used to drive 10-12 hours twice a year to visit my kids. It may sound stupid, but for me, it helped to break the trip up into pre-planned stop points, so you can rest, take a pee, fill up on gas, eat something, etc. It helps mentally if you are anticipating the next stop point, and not the end of the trip. Music is good, but if you are driving, try to avoid snacks and dvds! they can be very distracting.

A portable DVD player and lots of movies and tv shows on DVD. And the books on tape is an excellent suggestion as well.

Make sure you take a decent break every few hours. Will help prevent any nasties such as DVT.

Yeah don't drive with my wife. She rarely does any driving, complains when I stop, and complains when I slow down because I can barely see through the dirty windshield.
