I believe this is a "war" scene in Shakespeare right? Now is the hour of our discontent? These people are fed up and they are not taking any more, therefore they are ready "now" to take up arms or raise their pitchforks or whatever they had and fight the man/the power/the king to get the relief they so badly want. Do you remember in all the tellings of Robin Hood how everyone is in jail, and it looks very bleak for the whole story to end on a good note? It's sort of like that.
Everyone is oppressed, jailed, taxed beyond their means to survive on the limited income they had and what are they doing? Waiting for a miracle; Someone to change the balance of power so they can escape jail/ the oppression and/or pay their way out of debtor's prison and go back home. Robin Hood is the answer for that one. Tho, his friends get him out of jail a few times too.
Ok, so "nowis the hour" basically means we/the people have reached the breaking point and we aren't taking "it" anymore....whatever the problem or issue is.

It sounds as if the one who said it is a little bit exasperated.

Ususally one says, "Now is the time."

It means now is the time. As in now is the time to go to work, or now is the time to defend your country. It means it's the time to do something - whatever the speaker is referring to - now.
