oh, and it has to be testable

it has to be realistic

and to the first answerer, you can test if a fish is sad. And by testable, i mean measurable

and the test cant hurt any fish

Here's a few highly debated but easily tested questions. Some are obvious but this is based on some of the answers posted here:

Does salt kill freshwater ich?
Does a small amount of salt in the water kill fungus?
Does salt change the pH or hardness of the water?
Does carbon remove minerals such as salt from water?
Does the water in a tank that has carbon in the filter stay more clear than a tank with no carbon, provided they have the same fish and are fed the same?
Can you keep a betta with community fish?
Are piranha mean, will they attack a human hand in an aquarium (need a brave tester for this one lol)?
Which is faster, fishless cycling or cycling with a few small fish?

Those are just some I've seen in the last day or two. Most of these of course someone knows the correct answer, but even so it's fun and informative to do the research to test it for yourself. Some of course are easier to test than others. If you need help testing one of these feel free to email me. I won't give you the answer, even if I know it, but I will help you plan a totally unbiased test.

Sounds like your doing this for a report or science project. You could go with a water quality question. Water quality is easily measured with test kits as well.

How fast nitrates rise in a tank with a given amount of fish.
Effects of water quality on algae growth.
How effective plants are in an aquarium at removing nitrates from a tank.
What is the best type of filtration.
How much does carbon actually effect water quality.

An interesting one I have thought about was if fish can recognize certain people. You would need an assistant to help with that though. One person who always feeds the fish and another who never feeds them. Check for differances in behavor when each person goes up to the tank.

Well we know a few are out since we know salt doesn't kill ich tested and proven. MM is on the salt kick and will give you 100 bogus web pages. Email me if he does and I will give you 200 that prove him wrong. So let's just say Salt is out because every so called proof is disproven. Time to move on MM.

As far as any fish staying with another or if they are mean or not isn't a matter of prove or disprove just every fish is different with different attitudes. No science there.

So now you are down to no answers.
Here are a few:

Do fish see in color? Proven some fish do see in color while others don't. (Gold fish and Koi see in color as well as some salt water fish)

Does weather affect fish Proven high and low pressures do

Do fish feel pain (proven, yes fish feel pain)

Do fish reproduce for pleasure (nope, very few species do unlike dolphins and humans)

What does carbon do in a tank? (not much of anything. Pulls out all inorganic and organic's doesn't realize SALT is something that is supposed to stay!) Proven, carbon pulls out both.

Who can argue about fish the best (proven right here on Yahoo answers!)

Lol, here are a few.
-Do fish feel lonely when they are alone.
-Do fish sleep or do they just stop moving at night.
-Can fish feel sad when they lose a member of their group (in case of schooling fish)
-Can any type of fish be put together as long as they are fed properly and kept in good conditions.
