The easiest way to get to Uluru from Lismore is to drive or take a bus to Brisbane and then fly straight there. If you want to do it by car you have two routes. Lismore west to Tenterfield, south to Tamworth, west to Dubbo, west to Nyngan, through Cobar to Broken Hill, southwest to Peterborough, west to Port Augusta, north to Alice Springs, west to Uluru. The other route is north from Tenterfield to Toowoomba then northwest to Charleville, Longreach to Mt Isa then west to Three Ways and south to Alice Springs then west to Uluru. That's the longer way round. Allow the best part of a week to drive it either way.

No it's not a short distance.
The distance between Lismore NSW and Ayres Rock NT is approx 3100kms or 1926 miles.
