Yes. You must go through Customs no matter what mode of transportation you use, including by bus.

If you take a bus across the border, either direction, you must get off the bus, gather all your belongings and luggage, take it in to the checkpoint office, send it through the xray machines, answer questions about where you are going and why, then load all your things back onto the bus to continue the journey.

If there are any questions about your purposes, your baggage, or anything else, you may be questioned further. You can also be detained until the next bus comes through if there is reason to do so. Or if you are refused entry, then you are sent to the other side of the border to catch a bus back the other direction.

Of course there will be a security check at the border. No matter how you are travelling via bus, plane, train, or car. They will ask for your passport. You need to declare what you are bringing into the country.

Yes there will be. You'll go through security and immigration. They'll ask you why your visiting Canada, who your going to see if anyone and when your returning back to the USA. Usually it doesnt take long, as long as your honest with them.
