Google "amplifier" or "triode tube" or something similar and get a schematic of a simple one tube amplifier.

In the drawing on your link, several things are obvious:
You have connected the plate to ground - the plate must be a positive potential in relation to the cathode. The input must be routed to the control grid and the speaker connected to the secondary of the coupling or impedance matching transformer. A 12 volt supply without any step up is not going to supply you with sufficient B+ for the plate if you insist on using a vacuum tube.
Do not be discouraged. Study some schematics of simple working amplifiers and the process and the purpose of the different components will become clear.

No, it will not. The speaker doesn't connect to the plate. The transformer primary should be in series with the B+ supply for the plate, which is not shown. Input NEVER goes to the plate. Input goes to G1, or the control grid. You have left off any kind of grid leak resistor, and there is no cathode bias, with a by pass capacitor. The circuit, as it stands might produce a nice explosion as it stands, or just blow a fuse and leave you wondering why, except for answers given here. Where, or how did you dream this circuit up to begin with?

Ditto. WTF is that? You have the input driving the speaker and then going to the transistor, lol. That there is a smoke machine. Connect a 12V car battery to that sucker. Put your beer down first.
