There are a pair of regular customers at my work who are transvestites first and foremost, but also believe they are vampires (seriously, I'm not joking).

My issue is here that they come into my work during the day, and pretty obvious to everyone that vampires can't walk around during the day light. How should i approach this situation?

just start talking to them and tell them how you feel and want to ask some questions , no one is insulted if the questions are appropriate

Wow, that is a refreshingly new question!

Perhaps you could get into a conversation with them about ways in which traditions (or superstitions) about vampires are not real. For instance, you probably see their reflections in the glass of the store windows, which is not supposed to happen...you could ask if they get burned when they touch crosses or Holy water.

I would be interested to know what they say. Best wishes.

Just tell them you are looking into becoming a vampire and were wondering if they could help. Then ask away.

There is no such thing as vampires. There is no "situation". Just do your job with a polite smile.
