Well, maybe your butt is getting flatter because your're more sedentary, so you have less muscle. Butts can get rounder through workouts and such, so I imagine the opposite is true too when you don't work out.

No, sitting for a long time will not make your butt flatter, its just you who imagine your butt is getting flatter the whole time.

I don't think it make much difference, however sitting for long hours at a stretch does effect your lower back, so it is advised to get up and shake your legs or walk short distances to help blood circulation.
