The first city builder was Cain, who named the city after his son Enoch. Genesis 4:17 says, "Afterward Cain had intercourse with his wife and she became pregnant and gave birth to Enoch. Then he engaged in building a city and called the city’s name by the name of his son Enoch."

Cain the brother of Able took one of his sisters and went and built a city. He named the city after his first born son named Enoch, I believe.
