The prescription for your right and left eye could be completely different. If you wore you glasses upside down it would be like wearing someone elses glasses. Yes it could make a huge difference.

It does make a difference--
a) Left and right eyes are likely different prescriptions
b) Axis for astigmatism correction would be wrong
c) Optical centers would be off (not looking through center of lens)
d) Bifocal/varifocal upside-down
e) People would probably notice

waring glass upside down is OK i do it all the time but it is not OK when glasses are facing at you but i mean its fine is the nose pad or the arch is in the air

Besides being hard to keep them on your head since the eyepiece and earpiece are wrong, the bifocals would cause problems with sight.
