54th Massachusetts Volunteers is considered to be the first black unit, although there were others, and a few blacks here and there with regular units. They were originally offered less ($10) than white soldiers ($13), as they were not seen as full solidiers, and the unit voluntarily refused pay. After the Mass senate decided to give them equal pay, the unit still voluntarily refused it on principle. They did not take pay until equal pay was passed on a federal level.
Edit: It was an important step for the federal government in giving blacks equal treatment with whites.
Second Edit: The Buffalo soldiers were the first peacetime regiment, and were formed in 1866 by the fed gov't.. The 54th Mass. was formed in 1863, and were a unit, formed on the state level. The question was about the civil war, which was over in 1865, 1 year before the Buffalo Soldiers were formed. My answer is not incorrect.
All of the individual groups comprising the Buffalo Soldiers were formed 1866 or later, except the 25th infantry, which was not a black regiment. The 25th began accepting black soliers as part of the USCT (United States Colored Troops)which was formed May 22, 1863, 2 months after the 54th Massachusetts - March 1863. And, the actual FIRST in the war was the Louisiana Native Guard, but they were confederate.

Buffalo Soldiers is a nickname originally applied to the members of the U.S. 10th Cavalry Regiment of the United States Army by the native Apache indians, which was formed on September 21, 1866 at Fort Leavenworth, Kansas. The term eventually encompassed these units:
U.S. 9th Cavalry Regiment
U.S. 10th Cavalry Regiment
24th Infantry Regiment
25th Infantry Regiment
Although several black regiments were raised during the Civil War to fight alongside the Union Army (including the famous 54th Massachusetts Volunteer Infantry and the many United States Colored Troops Regiments), the "Buffalo Soldiers" were established by Congress as the first peacetime all-black regiments in the regular U.S. Army.
The Buffalo Soldiers were voluntary. The pay referenced above could be accurate but most even recruited, went as volunteers. I mainly know these facts as there is currently a group of motorcyclists that we ride with that are black...they are THE BUFFALO SOLDIERS they do a lot of good work and carry on the tradition.
Little note to be remembered:
On September 6, 2005, Mark Matthews, who was the oldest living Buffalo Soldier, died at the age of 111. He was buried at Arlington National Cemetery.
