If you are trying to test a new drug against a standard drug, here will be a good protocol:

The presenting population of patients can be alternatively assinged to two groups A and B. A can get the standard drug and B the new drug. A becomes the control and at the same time, gets the benefit of the standard treatment. Placebo effects are not present since both the groups have that part constant. And yes, the test should be double blind, i.e. the person administering the drug or the patient doesn't know whether A is getting the standard drug or the new drug.

If you are testing a new drug for a disease with no standard treatment available, then use any dummy placebo for one group and the new untested drug for the other group. The placebo receiving group will be a control group. The other features of the double blind trial will remain same.

You will be comparing the results of the two groups using some standard statistical tests. If you need more help, write separately or talk to a statistician in your lab. Best wishes.
