Greetings! One way people grieve God's spirit is by quenching His spirit. So many churches don't allow the Holy Spirit have full access to work in the services as He desire due to not wanting to offend people, not holding the service over a certain time, and by not allowing the gifts of the Spirit to function. They believe the Bible when it says despise not prophesyings, yet if someone were to prophesy... they'd be asked to leave and not return. We can quench the spirit by not doing what He would desire us to do. Many need a word of encouragement, but folks hold back due to fear and such. What grieves God is the sin of people who won't change their ways and allow Him to take control of their life. Hope this helps.

The particular passage you are refering to has to do with the people in Noah's time and says "5 Then the LORD saw that the wickedness of man was great on the earth, and that every intent of the thoughts of his heart was only evil continually. 6 The LORD was sorry that He had made man on the earth, and He was grieved in His heart."

I believe it is when we choose sin, over him. When we choose to seperate ourselves from his love, in order to fulfill the desires of our flesh, free from the Holy Spirits voice. I believe God grieves, not specifically because of our sins, but because he loves us, misses us, and doesn't want to see us hurt ourselves.
