When you say ‘popular’, that is a subjective and difficult thing to judge. The only real way we can determine this is to look at their sales figures. This includes not only comic books and movies, but action figures, lunch boxes, Underoos and all other merchandising.
On that basis, the two comic book characters who have sold the most are Tarzan and Superman. When you consider that Tarzan was a best seller long before modern comic books were invented, then you may have to discount him a bit. I think Batman is next, partly because when the Berlin Wall came down in 1989, the first person across was wearing a Batman T shirt, and everybody recognized him. I don’t know about the other 22.

Well, my favorites are Daredevil and Spider-Man, but you asked about the most popular. If my experiences in Marvel.com are anything to go by, here are the most popular Marvel heroes (in order):
Spider-Man
Wolverine
The Punisher (if you count him as a hero, which I actually don't, but most people do)
The Hulk
Captain America
The X-Men
Iron Man
The FF
Daredevil has a rather large following, largely because of the consistently superb quality of the comics since Frank Millar, but surprisingly little discussion goes on about him, so I'm not ranking him among the most popular.
I don't follow DC that much (except for Smallville), although of course I know that Superman and Batman are popular.
