What is the optimal way to keep the computer case cool, if you have 2 fans in the front and 2 fans in the back (none on the side), and the heatsink is positioned near the rear 2 fans? Which ones should blow air in and which ones out?

The idea is to pull cool air IN & push warm air OUT.
This said ... the optimal operation would be to have air blowing in at the front & blowing out at the rear.

This way (given your setup) the warm air from the heatsink is pushed out almost immediately & not dispersed further across the mainboard components.

Since the CPU (central processing unit) generates most of the heat in the case relative to the rest of the motherboard, you want to carry the heat away from the heatsink as quickly and efficiently as possible. Thus you want the fans closest to the heatsink blowing heated air out with the fans in the front blowing cool air in.
