That's a tough one. I'm going to say it's being able to grasp the concept of zero, and absence or nothingness, as opposed to presence.

knowledge is understanding the information when compared with the memory of the past accepted information.

Thus the role of knowledge in distinguishing 1 &0 is accepting or confirming that 1 is some thing and 0 is another thing as defined by the information acquired before. and that they are not the same.

Knowledge lets us remember the idea embedded in our mind which has existed before. It recalls the idea distinguishing 1 from 0 through experience and understanding. meaning to say, the idea/s distinguishing 1 from 0 has been totally impart of the mind, but due to occurence of experience at that point and time relating to the distinguishment of1 from 0 , knowledge recalls back that idea distinguishing 1 from 0 from the mind. Where there is experience there is knowledge and understanding.

Ans 1 Binary system

Ans 2 We have to know 1 as well as 0 to be able to distinguish between them. Unless we have knowledge of the two, we can not differentiate between the two.

The role of knowledge in distinguishing (not sure why you use that word) one from zero is from cognitive knowledge.

There can be NO knowledge of zero until there is a knowledge of one. And one is just a word to identify the amount of something.

Perhaps you could choose other words so that we can answer you more accurately.

Even a toad knows if there is 'something' or 'nothing'. Differentiating 1 from 0 requires sensory perception not knowledge if you refer to their literal meanings.
However if you mean quantifying 1 then yes I am of the opinion that a person requires a lot of knowledge to discern if there is truth, the whole truth and nothing but the truth.

0 and 1 are merely symbols - signs.
For distinguishing them by shape , knowledge is not necessary .But still there is a difference felt by us .
We give them values and there by make them different from each other .
The act of giving them their value we use our knowledge .
