Thou shall not kill is not a Bible quote, it is made up by people who are against capital punishment to sound like it came out of the Bible. Thou shall not murder is in the ten commandments in the Bible. It means to put murders to death, and don't commit murder.

The proper translation is "Thou shall do no murder".
If someone attacks you with a knife, and you defend yourself, which causes the attacker to die, then that is "killing".
But if you take a knife and stab some out of spite and that person dies, then that is "murder".

There is a difference between murder and killing. For example, if someone kicks in the your door, you have the right to protect yourself. If he dies, this is killing, not murder.
Murder is usually done for selfish reasons.
