This is a common problem with the application. The simplest test would be to restart your system and try again.
If this fails then I recommend that you redownload the application at http://www.apple.com/ipod/start/ don't worry your music library will stay intact assuming you choice this option in the installation process.
I would also ensure that the application is running as a process after you click the link. This will ensure that the icon you are clicking in not broken. I suggest you launch itunes from the shortcut in Start Menu or Widgets [Mac].

uninstall it
then restart your pc
install it again.. make sure you install it from www.itunes.com
