One theory is that by gaining enough knowledge, and using your mind enough, you can begin to use a higher percentage of your brain's total capacity (we typically use in the neighborhood of 10%). If one were able to learn enough to use even 50% of his/her brain, there is no telling what he/she could accomplish.

I was eight years old playing on a swing, when suddenly I was watching my body from up above somewhere swinging to and fro, and I thought it was the funniest thing I ever saw. It lasted a long time, but in reality, it was a mere minutes. I have never been able to do that again. I know there must be a scientific explanation for this because I don't believe in the immortal soul. It just happened. I don't know what caused it.

Can they? Yes
How? In the words of Bill and Ted "Be excellent to each other"
:)
Ok, so looking at these answers, I am wondering if you need to clarify what you are asking. Are you asking if humans can transcend the "human conditon" or are you asking if humans can transcend the physical realm for more cosmic experiences??? Please clarify...I took it in the sense of that we are our own worst enemy...
But then I gotta admit, the vagueness of the question, with a need for inference, got you a varied look at the people around you...
Starjumper: My guess would be that the rythm of the swinging lulled you in to a trance state, which allowed you to leave your body, but then whenever you have tried it again on purpose, you got too tense to do so.

Strictly speaking I believe human beings can overcome themselves more than transcend themselves. It depends on what you mean by transcendence. I don't believe that cosmic experiences that are out of the body are impossible but people can refine and sublimate their basic drives.

humans "transcend" themselves by negating the ego & stop pretending that they are separate from the rest of existence.
