A boxing right cross to the head is pretty brutal.
There's something I've seen in silat, where they punch at someone and grab their throat with their open hand (But with the full swing strength of a punch behind it). I don't practice silat myself, but I bet that hurts.

I would think that any quick bone break would be brutal.
Aside from the obvious "power punch/kick/knee" from a very powerfull individual, I think certain techniques are more brutal just by the basic mechanics of it.
guillotine- I think this one has the potential to do the most damage and hurt the person the most especially if you put enough pressure and aren't looking for a submission but to actually just damage your opponent (something you don't see in MMA events- this is one for you who say "the sport has rules", you want one of these thrown on you when there are none?).
RNC- what you gonna do when they come for you....
striking:
A "true" rabbit punch (punch to the back of the head) from the proper angle, that would mean from behind in an effort to deliver a killing blow in the way a rabbit is killed- hence the name (whats up with me and dead rabbits today?). Reaching around or attempting to simply strike someone in the back of the head on a shoot is not the kind of thing I'm talking about although it can be very damaging if it lands with enough force.
Attaks to the knee. particularly a "downward thrusting" against the inside of the knee.

Against a down opponent....jump high and land hard...
with a melee weapon? A good old bonk of the 13 foot staff in the nose.
Against any standing opponent....if you have some power....AND can land it....the move that will cancel an open-casket funeral...720 roundhouse ter the face or a neck crank that doesn't stop at the pop.
OK a realistic strike would be a kin geri to the solar plexus with double underhooks to cause death. IF you got the power to do it, of course. I'd imagine a merciless guillotine would do a cleaner job as would the taekwondo upward back hammerfist to the groin. Which is a VERY hard move to see coming or defend against because of the angle and range.
if you're looking for a simple move....grab the sucker's wrist with an opposite hand, move to the outside of his body, squat under his arm with your side turned toward him so the arm is over your shoulder......
.....and then stand up with all your might....

By far the most brutal martial art move is the Nattoh Umeboshii 27 snakes dimsum mak as taught by grand master of infinity Ashitda Dim of Koga Ryukododgi. The move starts by saying “this is by far the most deadly partial arts move on the planet and indeed the whole solar system, almost as deadly as the famously morbid death’s duck blow and dry from Alpha Zeee 333444 of Beta quadrant and guaranteed to cripple any would be attacker by way of split spleen.” Then adopt a left facing stance, take your hand up across your chest and breath in through your nose. Breathing out through your bottom step into a wide horse stance (primary dashi) and throw both hands across to your right in a blurry manner. Shout “Kiaih” and jump up in the air. Then do the “Dance of Deaf” and generally prance around oblivious to the music like your dad at your school disco. This move is guaranteed to kill even small children from 100 yards.

Elbow to the face is never a good thing.
Soccer kick to head while someone is down or worse, on all four is really bad.
Stomp on the face.
Any major joints lock is sick as well if follow through.
Knee or kick to someone kidney while they are down have a really nasty conquest.
Nothing that look cool and fancy are brutal. It's usually the most basic, crude, and barbarain technqiue that infect heavy damages.

I have seen several.... Be aware that some of them have to be performed on a crash test dummy or sth. like that. First of all all the Brazilian Jujitsu arm bars, leg and head locks are deadly if you choose to finish them all the way (head locks will most certainly cause immediate death) i especially enjoy the CRUCIFIX. Striking is brutal in any form but a Muay Thai knee to the face has a raw brutal power capable of killing the enemy instantly.... They tested the force and said it was equivalent to a 45 mph car crash with a concrete wall with no seat belts. And of course there are melee weapons like Katana - the ultimate killing tool... Any move with that weapon is extremely brutal.

I like this question. None of that Bruce Lee ninja bull ****.
Roundhouse kick. Cro Cop used this on Igor Vov. most brutal KO I've seen.
Straight right and left hook. Classic boxing punches when used in combination are devastating. A young Mike Tyson delivered some of the worst KO's ever using this.
Muay Thai clinch and knees to the head. W Silva used this on Rampage Jackson and left him lying unconscious on the ropes.
Ground and pound. Fedor Emelianeko has mastered this move, even when his opponents have him in their guard he can connect with numerous punches.

A back kick is pretty powerful. But I'd go with---I don't know the name, but it's real, it's in my patterns---the move that you gab someone by the neck and slam them into your knee. Oh, the nine block would hurt too---that breaks someones arm. Or ALL of the elbow moves I know. You know a kick to the groin would be the most deadly for guys. lol.
And yes, I can kick you butt.

That's easy enough....most brutal? I don't know if there's a MOST brutal per se, but there are techniques that are intended for posterior or anterior neck breaks, posterior spinal column breaks, and anything else that can decimate a joint....and grind the broken joint afterwards.
There are techniques designed for slow kills, and quick kills....the brutality aspect is based on perception I suppose.
The anatomy of these sort of things changes on so many levels depending on timing, distance, angling, whether I'm using a weapon/tool....and what that device happens to be.
Truly...there's no one answer for this, and I'm not about to give you directions on how to do these types of things.
A- you're not one of my students
B- this is the internet
Anyone who actually gives you examples or instructions of maiming/killing techniques online should be ashamed of themselves.

A walking stick/cane thrust to the throat.
Lots of momentum with a VERY small striking surface (1 or 2 sq. inches) targeting a very soft & vulnerable area!
