There is no such word as 'oo.' However, people may use it as an expression. 'Oh' is used in a reply a lot, for instance: "Oh, I thought that wasnt true." 'Oo,' on the other hand, is used to express special interest and amazement. Ex: "Ooo, that's pretty."

Oh: "Oh my goodness!" Or it is just commonly used to start out a phrase/reply: "Oh, that's okay."

Oo[o]: "Ooo, that is so cool!" Or it can be used to signify trouble: "Ooo, you're in huge trouble!"
