Here is a good list of all of the "largest" for aircraft in the world. There are many that can be called "largest" but in many different categories.

The plane in "Flightplan" is the E-474, a fictional plane that British Airways and Canadian Airways has attempted to actually purchase.

The E-474 looks like an A380 body, 747 wings and the original 787 cockpit design.

The Spruce Goose still has the largest span of any plane built, however it is shorter and much lighter than many modern aircraft.

As Matt says, the An-225 with a wing span of 291 feet and a MTOW of 1,400,000 is the biggest commercial aircraft in operation.

The second biggest would be the A380F at 271 feet and 1,300,000 pounds. The current pair of A380s are also 271 feet span but around 1,200,000 pounds.

sonicboom2885: While the Aan-225 may well have been developed for the military that doesn't make it a military aircraft. The Boeing 747 and Lockheed C-5 Galaxy both competed for a USAF heavy lift contract, the C-5 won but the contract and the bills the US government paid didn't make the 747 a military aircraft. There are plenty of examples of aircraft which were developed for the government but also saw commercial service, Shorts Belfast, Avro York, Avro Lincoln, DC-3 and even Concorde. The Royal Aircraft Establishment had a big role in Concorde and they were part of the MoD Procurement Executive and are my former employers. Was Concorde military?

Whoever said the A380 gets 80 miles to the gallon is WRONG!!! They are lucky if they get 1 Mile per gallon... they list the range at 8,000 nm... the fuel capacity is 81,890 gal... that means if they were getting 80 mpg, they would have a range of 6,551,200 miles... for comparison, thats 15 round trips to the moon and back... Just thought I'd throw that out there... additionally, the A380 wingspan is not 300 ft, or the size of a football field... it is 261ft. 8 in. That is almost 40 ft. or 13 yards shorter than a football field... The largest wingspan is the spruce goose, the miyra is the largest in terms of actual plane size though...

Matt is correct in stating that it is the Russian Antonov An-225 Myra as the largest aircraft in the world.
However, whoever it was that stated this is a commercial plane is incorrect. It was developed for the use of military transport, but has now been converted to general transport of goods.

The biggest passenger airplane in the world today is the Airbus A380, built jointly by Germany and France. It weighs 308 tons and takes 550 passengers. Its wingspan is bigger than a football field. It took 11 years to build and it cost $13,000,000,000 (13 billion) in development. It consumes one gallon of aviation fuel per passenger for every 80 miles of flight. Only recently it flew in New York, JFK Airport.
